Introduction to Mammalian Testis Histology
Mammalian testis histology deals with the microscopic structure of the two primary reproductive organs (gonads). The testis is responsible for spermatogenesis and secretion of androgens, mainly testosterone, whereas the ovary produces oocytes through oogenesis and secretes the steroid hormones estrogen and progesterone.
Histologically, the testis is characterized by numerous seminiferous tubules containing spermatogenic cells and supporting Sertoli cells, while the surrounding interstitial tissue contains hormone-secreting Leydig cells.
𝗚𝗲𝗻𝗲𝗿𝗮𝗹 𝗦𝘁𝗿𝘂𝗰𝘁𝘂𝗿𝗲 𝗮𝗻𝗱 𝗖𝗮𝗽𝘀𝘂𝗹𝗲𝘀
- The 𝘁𝗲𝘀𝘁𝗲𝘀 are paired male gonads responsible for producing 𝘀𝗽𝗲𝗿𝗺 and 𝗮𝗻𝗱𝗿𝗼𝗴𝗲𝗻𝘀 (e.g., testosterone).
- 𝗧𝘂𝗻𝗶𝗰𝗮 𝗔𝗹𝗯𝘂𝗴𝗶𝗻𝗲𝗮: A thick fibrous connective tissue capsule covering each testis. Its inner layer forms the 𝘁𝘂𝗻𝗶𝗰𝗮 𝘃𝗮𝘀𝗰𝘂𝗹𝗼𝘀𝗮, a loose connective tissue layer rich in blood vessels.
- 𝗟𝗼𝗯𝘂𝗹𝗲𝘀: Incomplete connective tissue septa project inward from the capsule, dividing each testis into roughly 𝟮𝟱𝟬 𝗹𝗼𝗯𝘂𝗹𝗲𝘀.
- 𝗠𝗲𝗱𝗶𝗮𝘀𝘁𝗶𝗻𝘂𝗺 𝗧𝗲𝘀𝘁𝗶𝘀: A posterior thickening of the tunica albuginea that projects inward, providing a passage for blood vessels, lymphatics, and genital ducts.
𝗦𝗲𝗺𝗶𝗻𝗶𝗳𝗲𝗿𝗼𝘂𝘀 𝗧𝘂𝗯𝘂𝗹𝗲𝘀
Each testicular lobule contains 1–4 seminiferous tubules surrounded by interstitial connective tissue containing Leydig cells.
𝗘𝗽𝗶𝘁𝗵𝗲𝗹𝗶𝘂𝗺: Lined by a complex stratified 𝘀𝗲𝗺𝗶𝗻𝗶𝗳𝗲𝗿𝗼𝘂𝘀 𝗲𝗽𝗶𝘁𝗵𝗲𝗹𝗶𝘂𝗺, made up of two basic cell populations:
- Spermatogenic cells undergo mitosis and meiosis at different stages of spermatogenesis and differentiate into mature spermatozoa: spermatogonia → primary spermatocytes → spermatids → sperm.
- 𝗦𝗲𝗿𝘁𝗼𝗹𝗶 𝗰𝗲𝗹𝗹𝘀 (𝗦𝘂𝘀𝘁𝗲𝗻𝘁𝗮𝗰𝘂𝗹𝗮𝗿 𝗰𝗲𝗹𝗹𝘀) tall, columnar, non-dividing supporting cells resting on the basal lamina, spanning the full thickness of the epithelium.
𝗧𝘂𝗻𝗶𝗰𝗮 p𝗿𝗼𝗽𝗿𝗶𝗮:A multilayered connective tissue around the basal lamina, consisting of 3–5 layers of 𝗺𝘆𝗼𝗶𝗱 𝗰𝗲𝗹𝗹𝘀 (peritubular contractile cells) whose rhythmic contractions help propel sperm and testicular fluid.
𝗦𝗲𝗿𝘁𝗼𝗹𝗶 𝗖𝗲𝗹𝗹𝘀: 𝗦𝘁𝗿𝘂𝗰𝘁𝘂𝗿𝗲 𝗮𝗻𝗱 𝗙𝘂𝗻𝗰𝘁𝗶𝗼𝗻
- 𝗨𝗹𝘁𝗿𝗮𝘀𝘁𝗿𝘂𝗰𝘁𝘂𝗿𝗲: Extensive smooth ER, well-developed rough ER, stacked 𝗮𝗻𝗻𝘂𝗹𝗮𝘁𝗲 𝗹𝗮𝗺𝗲𝗹𝗹𝗮𝗲, numerous mitochondria, Golgi apparatus, microtubules, and lipid droplets. The nucleus is ovoid or triangular, with a distinctive tripartite 𝗸𝗮𝗿𝘆𝗼𝘀𝗼𝗺𝗲 (an RNA-rich nucleolus flanked by DNA bodies).
- 𝗕𝗹𝗼𝗼𝗱-𝗧𝗲𝘀𝘁𝗶𝘀 𝗕𝗮𝗿𝗿𝗶𝗲𝗿: Formed by tight, parallel junctional complexes (over 50 fusion lines) between adjacent Sertoli cells, creating a physiological barrier that isolates developing germ cells from the bloodstream.
𝗙𝘂𝗻𝗰𝘁𝗶𝗼𝗻𝘀
- Act as "𝗻𝘂𝗿𝘀𝗲" 𝗰𝗲𝗹𝗹𝘀, facilitating nutrient/waste exchange and phagocytizing residual bodies during spermiogenesis.
- Secrete 𝗔𝗕𝗣 (𝗮𝗻𝗱𝗿𝗼𝗴𝗲𝗻-𝗯𝗶𝗻𝗱𝗶𝗻𝗴 𝗽𝗿𝗼𝘁𝗲𝗶𝗻) to concentrate testosterone within the tubule lumen. They also secrete 𝗶𝗻𝗵𝗶𝗯𝗶𝗻 to regulate the FSH feedback loop.
𝗟𝗲𝘆𝗱𝗶𝗴 𝗖𝗲𝗹𝗹𝘀 (𝗜𝗻𝘁𝗲𝗿𝘀𝘁𝗶𝘁𝗶𝗮𝗹 𝗖𝗲𝗹𝗹𝘀)
- Large, polygonal, 𝗮𝗰𝗶𝗱𝗼𝗽𝗵𝗶𝗹𝗶𝗰 cells located external to the myoid layer, within the interstitial stroma.
- Contain lipid droplets, lipochrome pigment, and distinctive refractile rod-shaped structures called 𝗰𝗿𝘆𝘀𝘁𝗮𝗹𝘀 𝗼𝗳 𝗥𝗲𝗶𝗻𝗸𝗲.
- They possess an elaborate 𝘀𝗺𝗼𝗼𝘁𝗵 𝗲𝗻𝗱𝗼𝗽𝗹𝗮𝘀𝗺𝗶𝗰 𝗿𝗲𝘁𝗶𝗰𝘂𝗹𝘂𝗺 (𝗦𝗘𝗥) and tubular-vesicular mitochondrial cristae, both specialized for 𝘁𝗲𝘀𝘁𝗼𝘀𝘁𝗲𝗿𝗼𝗻𝗲 𝘀𝘆𝗻𝘁𝗵𝗲𝘀𝗶𝘀.
